Full automation of high sensitivity SPE-LC-MS assay’s is now possible without compromising cycle time. With high speed On-line Solid Phase Extraction, PROSPEKT 2 sample clean-up can keep pace with state-of-the-art LC and LC-MS run times. Direct introduction of SPE eluate into the LC or LC-MS guarantees maximum sensitivity and eliminates manual sample transfers.

The Automatic Cartridge Exchange unit (ACE) has been designed with the “need for speed” in mind. Changing the disposable SPE cartridges is its main task and it does so quickly in order to reduce total cycle time. The optional second clamp opens the doors to parallel processing, further optimizing the total performance of the system.

pastilleA dedicated system for automated hyphenated SPE

The complete system for SPE with On-line elution consists of a PROSPEKT module, HPD and in most cases an Autosampler. The Sparklink software controls valve switching and automated SPE cartridge exchange with 200 cartridge capacity. The HPD (High Pressure Dispensor) selects and delivers SPE solvents (up to 24!) to the SPE cartridge, and the autosampler introduces the raw sample into the system.

The picture compares off-line SPE to on-line SPE as performed with PROSPEKT technology.
The key of on-line SPE is direct elution of the extract from the SPE cartridge into the LC system by the LC mobile phase. Several laborious handling steps are thus omitted, making on-line SPE much more efficient and providing superior analytical results.

The PROSPEKT-2 system can be configured in various ways to meet a large range of application requirements.

HYSPHERE SPE cartridges have been specially designed for on-line Solid Phase Extraction with PROSPEKT. Materials, geometry and packing procedures have been carefully optimized to ensurre maximum extraction performance and optimum compatibility with HPLC, LC-MS and GC requirements. Selection of sorbents for the HYSPHERE range is based on proven performance quality for on-line SPE and guaranteed control of physical and chemical characterisitics such as particle size distribution, absence of fines, batch-to-batch reproducibility etc. Microparticulate SPE sorbents (10µm) are used to provide optimum assay sensitivity.
